Why Semax as a nasal spray

Most peptides reach the market as injectables and are later tried intranasally. Semax went the other way. It was developed at the Russian Academy of Sciences as nasal drops, and the Russian clinical literature on it, stretching back to the 1980s, is overwhelmingly intranasal. When a paper describes what Semax does, it is usually describing what it does after going in through the nose.

The reasoning is anatomical. The upper nasal cavity sits directly beneath the brain, and the olfactory and trigeminal nerves that run through it offer peptides a route into the central nervous system that bypasses both the gut, which would digest them, and much of the blood-brain barrier. For a compound whose entire interest is its effect on the brain, that route is the point.

About Semax

Semax is a synthetic peptide originally developed in Russia as an analogue of a fragment of adrenocorticotropic hormone (ACTH 4–10), stripped of its hormonal activity while retaining its effects on the central nervous system. It is one of the most studied nootropic peptides in the research literature, with a body of work — mostly from Russian institutions — investigating its influence on brain-derived neurotrophic factor (BDNF) expression, attention, and neuroprotection following ischaemic events.

Research interest centres on sustained attention, working memory and mental stamina under cognitively demanding conditions. It has also been studied in the context of ADHD-type symptoms and post-stroke recovery, though these remain research contexts rather than approved clinical uses outside Russia.

BDNF after a single intranasal dose

Dolotov and colleagues (2006) found a single intranasal application in rats raised hippocampal BDNF protein about 1.4-fold and activated its receptor, trkB — the mechanism most Semax research leans on.

Learning and memory in animal models

The same work recorded more conditioned avoidance responses in treated animals, the behavioural half of the BDNF finding and the basis of the focus and memory positioning.

Neuroprotection

Russian clinical use centres on stroke and cerebral circulation. That work is largely published in Russian journals, which is why it is cited less often outside Russia than its volume would suggest.

Where the evidence is thin

Most of the Semax literature comes from a small number of Russian institutions, much of it in animals, and it has had little independent replication in Western trials. That is not the same as evidence against it, but it is a narrower base than the confidence of most online write-ups implies.

Using a nasal spray properly

    Prime the pump

    Before the first use, pump into the air until a fine, even mist comes out. Those first few sprays are not counted in the bottle total.

    Clear your nose

    Blow your nose gently first. A congested nose absorbs poorly and more of the spray runs straight out.

    Aim away from the middle

    Tilt your head slightly forward and point the nozzle toward the outer wall of the nostril, not the septum.

  4. 4

    Breathe in gently

    Press once while breathing in softly through the nose. A hard sniff pulls the spray down the throat instead of leaving it in the nose.

    Alternate nostrils

    Between sprays, alternate nostrils. That spreads the dose over more of the nasal lining.

Storage

Keep the bottle upright with the cap on, and in the fridge once opened. The amber glass protects the solution from light, but not from heat: don’t leave it in a parked car or a sunny window, especially in a South African summer. Don’t freeze it, and don’t share a nozzle between people.
