About Retatrutide
Retatrutide is a triple agonist: it acts on the GLP-1, GIP and glucagon receptors at once. Semaglutide works on one of those, tirzepatide on two, and the trial results have followed that progression — each additional receptor has produced larger effects on body weight than the generation before it. Retatrutide is the current end of that line and its phase 2 results were, by some margin, the largest seen in this class.
The glucagon receptor is what makes it structurally different from tirzepatide rather than simply stronger. Glucagon receptor agonism raises energy expenditure, which means Retatrutide is not only suppressing intake the way a pure GLP-1 does — it is acting on both sides of the balance. It is also the newest of the three, with correspondingly less long-term data behind it, which is a real consideration and not a marketing footnote.
