Why DSIP as a nasal spray

Delta Sleep-Inducing Peptide was isolated in 1977 from the blood of rabbits in induced sleep, and named for the slow-wave (delta) sleep it seemed to promote. Much of the early human work gave it intravenously, including trials in insomnia and in shifted sleep-wake cycles.

The spray is a practical format rather than one with a large literature of its own: DSIP is a small peptide, and intranasal delivery is the needle-free alternative to the injections the older studies used. That makes the spray convenient, but it also means the intranasal-specific evidence is thinner here than for Semax or oxytocin.

About DSIP

Delta Sleep-Inducing Peptide (DSIP) was first isolated in the 1970s during studies on sleep induction, and named for its association with delta-wave (slow-wave) sleep activity on EEG. It has since been studied for a potential role in stress regulation, pain modulation and normalising disrupted sleep-wake cycles.

Despite the name, DSIP’s direct sleep-inducing effect in humans has produced mixed results across studies. Researchers now investigate it more broadly as a stress-and-circadian-regulation peptide than as a straightforward sedative — its research interest lies in sleep architecture and recovery rather than sedation.

Sleep architecture

Research interest is in the structure of sleep — the share and quality of slow-wave sleep — rather than simply falling asleep faster.

Shifted sleep-wake cycles

Schneider-Helmert (1987) studied DSIP in phase-shifted insomnia, where the problem is timing rather than the ability to sleep.

Stress regulation

Researchers now describe DSIP more as a stress and circadian regulation peptide than a sedative, which is how we position it.

Where the evidence is thin

A 2006 review in the Journal of Neurochemistry called DSIP "a still unresolved riddle": its gene and receptor have never been identified, and the case for it as a natural sleep factor is weak. Some synthetic DSIP analogues promoted slow-wave sleep in animals where DSIP itself did not. Go in expecting research into sleep quality and recovery, not a sleeping pill.

Using a nasal spray properly

    Prime the pump

    Before the first use, pump into the air until a fine, even mist comes out. Those first few sprays are not counted in the bottle total.

    Clear your nose

    Blow your nose gently first. A congested nose absorbs poorly and more of the spray runs straight out.

  3. 3

    Aim away from the middle

    Tilt your head slightly forward and point the nozzle toward the outer wall of the nostril, not the septum.

  4. 4

    Breathe in gently

    Press once while breathing in softly through the nose. A hard sniff pulls the spray down the throat instead of leaving it in the nose.

    Alternate nostrils

    Between sprays, alternate nostrils. That spreads the dose over more of the nasal lining.

Storage

Keep the bottle upright with the cap on, and in the fridge once opened. The amber glass protects the solution from light, but not from heat: don’t leave it in a parked car or a sunny window, especially in a South African summer. Don’t freeze it, and don’t share a nozzle between people.
