The GLOW Stack

The GLOW Stack is a beginner-friendly introduction to peptides for skin regeneration and anti-aging. Learn which compounds are most relevant, how they work together and how to start safely.

The anti-aging skincare industry generates billions every year on the promise of younger-looking skin. Most of it delivers modest results at best. The active ingredients in the vast majority of mainstream anti-aging products, peptide fragments, retinoids, antioxidants, are genuinely useful in principle but often delivered in concentrations too low, in vehicles with poor penetration, or without the complementary compounds needed to produce meaningful change.

The GLOW Stack is a more targeted approach. It draws on the compounds with the strongest research profiles for skin regeneration and combines them in a way that addresses the multiple biological mechanisms behind skin aging simultaneously.

This article explains what skin aging actually involves at a cellular level, which peptides are most relevant, how they work together and how to get started.

What Actually Causes Skin to Age?

Before choosing compounds, it helps to understand the mechanisms you are trying to address. Skin aging is not a single process. Several distinct biological changes happen simultaneously and reinforce each other.

Collagen loss and disorganisation
Collagen is the primary structural protein in skin. It provides firmness, tensile strength and the scaffold on which skin sits. From the mid-twenties, collagen production slows by approximately 1% per year. Beyond just declining quantity, existing collagen becomes progressively more cross-linked and disorganised, making it stiffer and less functional. The result is the loss of firmness and the development of lines and folds that deepens over decades.

Elastin degradation
Elastin is the protein that gives skin its ability to spring back after being stretched or compressed. Like collagen, elastin production slows significantly with age, and exposure to UV radiation accelerates the breakdown of existing elastin fibres. Loss of elastin is one of the primary causes of the sagging, crepey quality that characterises aging skin.

Reduced hyaluronic acid
Hyaluronic acid is a naturally occurring molecule that holds water in the dermis, keeping skin plump and hydrated from within. Its production declines significantly with age, which is a major contributor to the thinner, drier quality of older skin.

Declining cellular turnover
Young skin replaces its outermost layer (the epidermis) approximately every 28 days. As we age, this turnover slows, and dead skin cells accumulate longer on the surface, contributing to dullness and uneven texture.

Oxidative stress and inflammation
Chronic exposure to UV radiation, pollution, stress and metabolic byproducts generates reactive oxygen species (free radicals) in skin tissue. These damage collagen, elastin and DNA in skin cells. Chronic low-grade inflammation, driven partly by senescent cells in the dermis, compounds this damage over time.

Reduced angiogenesis and microcirculation
The capillary networks that supply the dermis with nutrients and oxygen become less dense with age. This reduced perfusion limits the supply of everything the skin needs for repair and renewal.

The Core Compounds of the GLOW Stack

The GLOW Stack is built around compounds that each address specific mechanisms of skin aging.

GHK-Cu (Copper Peptide)
The anchor compound of the stack. As covered in detail in our GHK-Cu article, this tripeptide stimulates collagen and elastin synthesis, supports hyaluronic acid production, promotes angiogenesis in the dermis, activates enzymes that clear old disorganised collagen and exerts significant antioxidant and anti-inflammatory effects.

GHK-Cu addresses more mechanisms of skin aging simultaneously than almost any other compound, and its clinical evidence base for skin applications is among the strongest available for any cosmetic peptide. It can be applied topically for localised skin effects or administered subcutaneously for systemic effects alongside skin benefits.

Epithalon
While Epithalon's primary longevity mechanism relates to telomere maintenance and telomerase activation, its relevance to skin is significant. Telomere shortening in skin cells, particularly fibroblasts (which produce collagen and elastin), is one of the reasons skin cell function deteriorates with age. Epithalon's support for cellular longevity in fibroblasts contributes to maintaining their ability to produce structural proteins over time.

Epithalon also supports melatonin production (through its pineal gland associations), and melatonin is one of the most potent antioxidants active in skin tissue. Its anti-inflammatory effects add another mechanism relevant to the inflammatory component of skin aging.

BPC-157
BPC-157 is primarily known as a recovery peptide, but its wound healing research is directly relevant to skin regeneration. Its promotion of angiogenesis improves the microcirculation that aging skin loses. Its anti-inflammatory properties address a key driver of dermal aging. Some research also suggests BPC-157 influences the production of growth factors relevant to skin repair.

For people with specific skin concerns like scarring, stretch marks or localised areas of significant damage, BPC-157 is a useful addition to the stack.

NAD+ Precursors (NMN or NR)
As discussed in our NAD+ article, declining NAD+ reduces sirtuin activity, which in turn impairs DNA repair and cellular stress responses in skin cells. Sirtuin activity, particularly SIRT1, has been specifically studied in skin tissue and found to play a role in UV damage repair and the regulation of senescence in skin cells.

Supporting NAD+ levels is not a skin-specific intervention, but the skin-relevant effects of maintaining sirtuin activity and reducing cellular senescence in dermal tissue make NAD+ support a logical addition to a skin regeneration protocol.

How to Structure the GLOW Stack

The stack can be approached in layers, from accessible to more advanced.

Starting point (topical focus, minimal injections):
Begin with a high-quality topical GHK-Cu serum applied daily. Add an NMN or NR supplement orally. This combination addresses collagen stimulation, antioxidant protection and NAD+-dependent cellular repair without any injections. It is the most accessible entry point and appropriate for someone new to the space.

Intermediate (adding subcutaneous peptides):
Add subcutaneous GHK-Cu injections (1 to 2mg per day) to elevate systemic GHK-Cu levels beyond what topical application delivers. Incorporate a course of Epithalon (5 to 10mg daily for 10 to 20 days, one to two times per year). Continue the topical GHK-Cu and NAD+ supplementation.

Advanced (full protocol):
The complete stack adds BPC-157 (250 to 500mcg per day subcutaneously) for its angiogenic and tissue repair contributions, particularly if scarring, specific skin damage or poor microcirculation is a concern. This level of the stack involves multiple compounds and requires careful protocol design and quality sourcing.

Supporting the Stack from the Outside

The most sophisticated peptide protocol will be significantly undermined if basic skin protection and lifestyle factors are ignored. The GLOW Stack works best when combined with:

Consistent sun protection: UV radiation is the single largest extrinsic driver of skin aging. Mineral SPF applied daily is non-negotiable.

Adequate sleep: Skin repair happens primarily during sleep, driven by growth hormone release and reduced cortisol. Poor sleep quality has measurable effects on skin appearance and healing.

Nutrition: Collagen synthesis requires vitamin C as a cofactor. Antioxidants from food support the same pathways that GHK-Cu protects chemically. Adequate protein provides the amino acid building blocks for structural skin proteins.

Hydration: Hyaluronic acid binds water in the dermis, but there needs to be adequate water available to bind. Chronic mild dehydration contributes to the thinning and reduced plumpness of aging skin.

Realistic Expectations

Skin aging takes decades to develop and cannot be reversed in weeks. The GLOW Stack, used consistently, is likely to produce:

  • Gradual improvement in skin firmness and texture over months of use
  • Reduced inflammation and more even skin tone
  • Improved skin resilience and healing speed for minor damage
  • Over the longer term, a slower rate of structural deterioration compared to doing nothing
